Mouse IL-33

IL33 is a cytokine that plays a crucial role in immune responses, particularly in the context of allergic reactions and tissue injury.

Key Roles:

  • Receptor Interaction: IL33 binds to the IL1RL1/ST2 receptor, activating NF-kappa-B and MAPK signaling pathways in target cells, leading to various immune responses.
  • Th2 Cell Maturation: It is involved in the maturation of T-helper type 2 (Th2) cells and promotes the secretion of Th2-associated cytokines, enhancing allergic and anti-parasitic responses.
  • Activation of Immune Cells: IL33 activates mast cells, basophils, eosinophils, and natural killer (NK) cells, contributing to the orchestration of the immune response.
  • Macrophage Polarization: It enhances the polarization of alternatively activated macrophages, which are involved in resolving inflammation.
  • Chemoattraction: IL33 acts as a chemoattractant for Th2 cells and may function as an "alarmin," amplifying immune responses during tissue injury.
  • Mitochondrial Rewiring: It induces mitochondrial changes that reduce reactive oxygen species (ROS) production while maintaining the Krebs cycle, facilitating the differentiation of macrophages towards an anti-inflammatory phenotype.
  • Nuclear Role: In its uncleaved form, IL33 is constitutively expressed in quiescent endothelial cells, acting as a chromatin-associated factor that represses transcription by sequestering NF-kappaB/RELA.

IL33 is significant for its dual roles in promoting Th2 responses and acting as a damage-associated molecular pattern (DAMP) during tissue injury, linking innate and adaptive immunity.
