IL-36G binds to the IL1RL2/IL-36R receptor, activating NF-kappa-B and MAPK signaling pathways, similar to the IL-1 system. It is involved in local inflammatory responses, particularly in epithelial tissues. 

IL-36G enhances skin inflammatory responses by acting on keratinocytes and dendritic cells, promoting tissue infiltration, cell maturation, and proliferation. It induces chemokines (e.g., CCL3, CCL4, CCL5) and pro-inflammatory factors like TNF-alpha and S100A7/psoriasin. Additionally, IL-36G activates lung fibroblasts, increasing IL-8 and CCL20 expression, and may play a role in the innate immune response to fungal pathogens like Aspergillus fumigatus.
