Secreted mainly by activated T-lymphocytes, mast cells, and osteoblasts, IL-3 regulates the production and differentiation of hematopoietic progenitor cells into specific lineages. It activates mature basophils, eosinophils, and monocytes, and plays a role in neural cell proliferation and survival. IL-3 is also involved in bone homeostasis by inhibiting osteoclast differentiation through NF-kappa-B inhibition. Its effects are mediated via a receptor composed of IL3RA and IL3RB, activating JAK2 and triggering a STAT5 transcriptional program, while also supporting cell survival under oxidative stress through PI3K/AKT and ERK pathways.
